Why does the room over the garage heat up first and cool last?

Will closing the register or blocking vents fix the temperature in that room?

Closing the register does not fix an unbalanced duct run to a room this far out. The ducts were never sized or sealed well enough to reach that distant space, so you just starve the rest of the house without ever making this room comfortable. A dedicated system is the way to stop fighting it.

No. A ductless mini split does not use the existing ducts at all. The indoor unit goes on the wall in the room over the garage and a small line set connects to the outdoor unit, so we solve the problem at its source without tearing into the garage ceiling or trying to rebalance the old run.
